high ribo % can be indicative of low quality cells. Depends on your tissue and organism

I would do some clustering and see if there are specific clusters with very high % ribo, these might be problematic

Heya,

I have quite some experience in analysing scRNA datasets, however I never worked with time course data.

For an upcoming project I will have a mouse disease model with samples from different time points.

What are some algorithms / papers to look at that model cell composition or gene trajectories over time?

What has your experience been in "integrating" datasets in a setting like that?

Using Pseudobulk Approach for Identifying Marker Genes Within a Single Condition

Pseudobulking of cell types to find markers seems like a good idea to me. The resulting p-values will be better than Wilcoxon dervied ones at least. Just make sure to filter for genes that are expressed in a high % of cells to get good results.

Sowohl Wiso Steuer und das Finanzamt meinen ich müsste was nachzahlen, also hatte ich das gemacht.

Habe einen ETF mit Freistellungsauftrag unter der 1000 € Grenze, da hat sich zum Vorjahr auch nix geändert

Hi,

habe gerade die zweite Steuererklärung meines Lebens gemacht.

Ich war etwas erstaunt dass das Steuerprogramm anders als im Vorjahr eine Nachzahlung von 41 € ausgespuckt hat.

Woran kann das liegen? Meine Steuersituation ist super simpel, ich habe lediglich im Oktober eine Gehaltserhöhung bekommen. Kann das die Ursache sein? Im Festsetzungsbescheid steht lediglich "Einkommensteuer zu wenig gezahlt."

Was wäre passiert wenn ich keine Steuererklärung abgegeben hätte und wäre ich dazu verpflichted gewesen?

Erhöhung von 65% auf 85% in Oktober

Was mir bei der Debatte etwas fehlt. Ja die CO2 Bilanz ist besser als Vorher, aber wäre sie nicht noch besser mit den AKWs und noch weniger Kohle?

Oder ginge das mit den veralteten Kraftwerken auf keinen Fall weiter?

I think it is preference. IIRC Edger is supposed to be slightly more conservative compared to Deseq2. I use PyDeseq2 to spare myself from firing up R

You could also give this recent method a try:
