About Ultimate Popping YouTube Channel:
Ultimate Popping publishes all new original content 3 times a week: every Monday, Wednesday, and Friday at 12:00 noon (MST). I’ve been delivering this all new original content for months without fail, and I don’t plan to stop anytime soon!
The “Ultimate Popping” YouTube Channel is dedicated to all things popping. I make videos of popping blackheads, whiteheads, cysts, sebaceous cysts, and all other types of acne and skin blemishes. Another thing that you will see me making popping videos of is sebaceous filaments. My face – specifically my nose and cheeks – have some kind of insane remarkable ability produce huge amounts of sebum in the form of sebaceous filaments. With the right tool – which can be just about anything – I can pop unreal amounts of pus out of my nose and cheeks. I haven’t decided yet if this is a curse or a blessing, but for now, I am really enjoying making videos of popping all of them.
In addition to popping, I also make a lot of videos of plucking (tweezing). I have two pairs of Tweezerman tweezers: a regular blunt ended pair and an ingrown hair pair that has an extremely sharp end that helps to get ingrown hairs started when they are under the skin. When I’ve recently shaved, I can find many ingrown hairs on my face and neck, as well as occasionally on my body. I enjoy tweezing and plucking these and making videos of it. Furthermore, someone I know has a hairy mole that grows back its long, thick, black hairs in a shockingly short amount of time! Thus, I get to film the removal of the hairs from this hairy mole every so often. It’s sort of like a game I play to see how many hairs I can pluck without breaking any of the hairs.
I also make videos of removing cases of what is known as ‘Pili Multigemini.’ Pili Multigemini is when a hair follicle is malformed and several hairs are growing out of the same hair follicle. When you see these growing on your face or body, it looks sort of like a pitchfork or some talons emerging from a single hair follicle. Often times it looks like its multiple hairs, but it is actually one hair follicle with multiple hairs or multiple shafts breaking off from the hair.
One of the questions I am most often asked is what type of instrument I use in my videos. The instrument I use is called a comedone extractor, also known as a ‘comedone remover.’ I made a video review of the actual comedone removers I use, and a link to the product on Amazon is provided in the video (see links provided below). My videos are recorded using a basic USB microscope camera.

I was using my Tweezerman ingrown hair tweezers to remove ingrown hairs and hair follicle malformations such as pili multigemini, and I noticed a peculiar looking little piece of stubble. It was not an ingrown hair or a pili multigemini, but the base of the hair seemed to be a lot thicker than all the other hairs. I decided to go ahead and pluck it, and I was totally blown away by how FAT and HUGE the hair follicle was that came out with it! It was completely intact and bulbous.
As soon as I watched this footage, it immediately brought to mind my years as a child playing Super Mario Bros 2 on the Nintendo Entertainment System when Mario pulls on a little root and a massive huge enourmous carrot or potato comes out of the ground!

Almost all of my hair is Pili Multigemini follicles, which is a condition where more than one hair grows per follicle, they can have up to 6 hairs in one follicle (it could be more but the highest I counted was 6).
It was pretty hard to record this using the phone as my eye to see the tweezers, because of the light I had to position the phone in my belly so you migh hear some breathing aswell.

Hair follicle
A hair follicle is a mammalian skin organ that produces hair. Hair production occurs in phases, including growth (anagen), cessation (catagen), and rest (telogen) phases. Stem cells are responsible for hair production. The shape of the hair follicle has an effect on the hair shape and texture of the individual's hair.
Structure
Papilla
The papilla is a large structure at the base of the hair follicle. The papilla is made up mainly of connective tissue and a capillary loop. Cell division in the papilla is either rare or non-existent.
Matrix
Around the papilla is the hair matrix.
Root sheath
The root sheath is composed of an external and internal root sheath. The external root sheath appears empty with cuboid cells when stained with H&E stain. The internal root sheath is composed of three layers, Henle's layer, Huxley's layer, and an internal cuticle that is continuous with the outermost layer of the hair fiber.
Bulge
The bulge is located in the outer root sheath at the insertion point of the arrector pili muscle. It houses several types of stem cells, which supply the entire hair follicle with new cells, and take part in healing the epidermis after a wound. Stem cells express the marker LGR5+ in vivo.

Hair

Almost all of my hair is Pili Multigemini follicles, which is a condition where more than one hair grows per follicle, they can have up to 6 hairs in one follicle (it could be more but the highest I counted was 6).
It was pretty hard to record this using the phone as my eye to see the tweezers, because of the light I had to position the phone in my belly so you migh hear some breathing aswell.

The instrument I use is called a comedone extractor, also known as a ‘comedone remover.’ I made a video review of the actual comedone removers I use, and a link to the product on Amazon is provided in the video (see links provided below). My videos are recorded using a basic USB microscope camera.

I have two pairs of Tweezerman tweezers: a regular blunt ended pair and an ingrown hair pair that has an extremely sharp end that helps to get ingrown hairs started when they are under the skin. When I’ve recently shaved, I can find many ingrown hairs on my face and neck, as well as occasionally on my body. I enjoy tweezing and plucking these and making videos of it. Furthermore, someone I know has a hairy mole that grows back its long, thick, black hairs in a shockingly short amount of time! Thus, I get to film the removal of the hairs from this hairy mole every so often. It’s sort of like a game I play to see how many hairs I can pluck without breaking any of the hairs.
I also make videos of removing cases of what is known as ‘Pili Multigemini.’ Pili Multigemini is when a hair follicle is malformed and several hairs are growing out of the same hair follicle. When you see these growing on your face or body, it looks sort of like a pitchfork or some talons emerging from a single hair follicle. Often times it looks like its multiple hairs, but it is actually one hair follicle with multiple hairs or multiple shafts breaking off from the hair.
One of the questions I am most often asked is what type of instrument I use in my videos. The instrument I use is called a comedone extractor, also known as a ‘comedone remover.’ I made a video review of the actual comedone removers I use, and a link to the product on Amazon is provided in the video (see links provided below). My videos are recorded using a basic USB microscope camera.

I was using my Tweezerman ingrown hair tweezers to remove ingrown hairs and hair follicle malformations such as pili multigemini, and I noticed a peculiar looking little piece of stubble. A Pilar Cyst, aka a trichilemmal cyst, an isthmus-catagen cyst, or a “wen”, looks identical on examination to the more common epidermoid cyst, except that 90% of them occur on the scalp. Interestingly, hiPSC-derived keratinocyte precursors obtained in the middle of the induction toward adult-type keratinocytes better communicated with human dermal papilla (DP) cells in co-culture and, more importantly, contribute to hair follicle structure in in vivo HF reconstitution experiment.
These findings suggested that hiPSCs may be used to produce epithelial cells more receptive to hair inductive dermal signals. For trichogenic dermal cell preparation, direct induction of hiPSCs into DP cells may be difficult.
Recently, we successfully generated MSCs from hiPSCs. Importantly, hiPSC-derived MSCs (hiPSC-MSCs) contained larger number of highly proliferative and multipotent CD271+CD90+ subset than human bone marrow-derived MSCs. When exposed to a culture condition maintains/restores human DP cell properties, CD271+CD90+ but not CD271-CD90+ hiPSC-MSCs up-regulated DP signature genes. In addition, when co-cultured with keratinocytes, CD271+CD90+hiPSC-MSCs bidirectionally communicate with keratinocytes to increase HF related gene expression in keratinocytes and further up-regulated DP biomarkers and the efficiency of this crosstalk was greater than that between cultured human keratinocytes and DP cells.
These observations, together with the fact that melanocytes can be generated from hiPSCs, support that hiPSCs can be a promising cell source for human HF regeneration.

Summary:
Use of human induced pluripotent cells (hiPSCs) for tissue engineering is generally considered to be beneficial. In case of the skin, putative epithelial and dermal stem cell populations exhibiting stem cell properties have been isolated. Thus, the advantage of hiPSCs as the material for bioengineering of the skin and its appendages may seem to be moderate. Yet, we still see hiPSCs as a promising material for human hair follicle (HF) regeneration.
In this talk, recent examples of applying hiPSC for human HF bioengineering will be presented. hiPSCs can be programmed into keratinocyte lineages. A Pilar Cyst, aka a trichilemmal cyst, an isthmus-catagen cyst, or a “wen”, looks identical on examination to the more common epidermoid cyst, except that 90% of them occur on the scalp. They tend to be more mobile and firmer than epidermoid cysts, and are derived from a different part of the hair follicle/skin. The internal content tend to be more “homogenous”/ smooth (looks different under the microscope), but it is also a benign growth. It is not uncommon to have multiple pilar cysts on the scalp and to have other family members to have these cysts (tend to run in families). Treatment is for cosmetic reasons usually (people don’t like the appearance of them), and excision with removal of the cyst and it’s contents is curative.
