Starch is a component in many foods. When iodine is added to starch it turns blue. That is why, in the experiment, the solutions in both beakers turn blue. On addition of saliva which contains an amylase, a starch digesting enzyme (chemical compound that catalyses the breakdown of starch); the starch gradually converts to sugar. Once the starch is broken down, the blue colour disappears. It is important to know that the salivary amylase requires an alkaline medium to act.

c) Pour 10 ml of amylase solution
d) Add 3-4 drops of Iodine solution
e) Place in a test tube rack

e) Place in a water bath between 37-42 degrees celcius

Simple and easy experiment to demonstrate the iodine with starch reaction!
For this test you will need:
• Two test tubes
• Soluble starch powder
• Iodine solution
• Water
• Dropper
Procedure:
• Put some starch powder into a test tube and fill the test tube with water.
• Mix the starch in the test tube well until the starch dissolves in the water.
• Fill the other test tube with normal water.
• Place both the test tubes in a test tube stand.
• Using a dropper take iodine solution.
• Put some drops in each test tube.
• Observe that the test tube with starch solution turns to purple black color.
• The other test tube with normal water retains the color of iodine i.e orange or yellow.
Explanation:
Starch is a mixture of amylose and amylopectin, which are different forms of glucose/starch.
Amylose in starch is responsible for the formation of a deep blue black color.
Amylase is long polymer chains of glucose units connected by an alpha acetal linkage and looks much like a coiled spring.
However iodine is a potassium iodide reagent and it is not very soluble in water.
So, iodine is prepared by dissolving it in water in an aqueous solution of potassium iodide. This results in a linear tri-iodide ion (I3−) complex in iodine which is soluble.
This tri-iodide ion (I3−) slips into the coil of the starch causing an intense or deep blue-black color.

Iodine
Iodine is a chemical element with symbol I and atomic number 53. The name is from Greek ἰοειδής ioeidēs, meaning violet or purple, due to the color of iodine vapor.
Iodine and its compounds are primarily used in nutrition, and industrially in the production of acetic acid and certain polymers. Iodine's relatively high atomic number, low toxicity, and ease of attachment to organic compounds have made it a part of many X-ray contrast materials in modern medicine. Iodine has only one stable isotope. Iodine radioisotopes, such as 131I, are also used in medical applications.
Iodine is found on Earth mainly as the highly water-soluble iodide ion I−, which concentrates it in oceans and brine pools. Like the other halogens, free iodine occurs mainly as a diatomic molecule I2, and then only momentarily after being oxidized from iodide by an oxidant like free oxygen. In the universe and on Earth, iodine's high atomic number makes it a relatively rare element. Its presence in ocean water has given it a role in biology. It is the heaviest essential element used widely by life in biological functions (only tungsten, employed in enzymes by a few species of bacteria, is heavier). Iodine's rarity in many soils, due to initial low abundance as a crust-element, and also leaching of soluble iodide by rainwater, has led to many deficiency problems in land animals and inland human populations. Iodine deficiency affects about two billion people and is the leading preventable cause of intellectual disabilities.

Tincture of iodine
Tincture of iodine or iodine tincture is an antiseptic, it is also called weak iodine solution. It is usually 2–7% elemental iodine, along with potassium iodide or sodium iodide, dissolved in a mixture of ethanol and water. Tincture solutions are characterized by the presence of alcohol. It was used from 1908 in pre-operative skin preparation by surgeon Antonio Grossich.
USP formulas
USP Tincture of Iodine is defined in the U.S. National Formulary (NF) as containing in each 100 mL, 1.8 to 2.2 grams of elemental iodine, and 2.1 to 2.6 grams of sodium iodide. Alcohol is 50 ml and the balance is purified water. This "2% free iodine" solution provides about one mg of free iodine per drop.
USP Strong Iodine Tincture is defined in the NF as containing in each 100 mL, 6.8 to 7.5 gram of iodine, and 4.7 to 5.5 gram of potassium iodide. Purified water is 50 mL and the balance is alcohol. This 7% tincture solution is about 3.5 times more concentrated than USP 2% tincture.
As in the case of Lugol's iodine, the role of iodide in the solution is to increase the solubility of the elemental iodine, by turning it to the soluble triiodide anion I3−. However, since iodine has moderate solubility in ethanol, it is also assisted by this solvent directly. Lugol's iodine, by contrast, has no alcohol, and has twice the amount of iodide as elemental iodine.

For this test you will need:
• Two test tubes
• Soluble starch powder
• Iodine solution
• Water
• Dropper
Procedure:
• Put some starch powder into a test tube and fill the test tube with water.
• Mix the starch in the test tube well until the starch dissolves in the water.
• Fill the other test tube with normal water.
• Place both the test tubes in a test tube stand.
• Using a dropper take iodine solution.
• Put some drops in each test tube.
• Observe that the test tube with starch solution turns to purple black color.
• The other test tube with normal water retains the color of iodine i.e orange or yellow.
Explanation:
Starch is a mixture of amylose and amylopectin, which are different forms of glucose/starch.
Amylose in starch is responsible for the formation of a deep blue black color.
Amylase is long polymer chains of glucose units connected by an alpha acetal linkage and looks much like a coiled spring.
However iodine is a potassium iodide reagent and it is not very soluble in water.
So, iodine is prepared by dissolving it in water in an aqueous solution of potassium iodide. This results in a linear tri-iodide ion (I3−) complex in iodine which is soluble.
This tri-iodide ion (I3−) slips into the coil of the starch causing an intense or deep blue-black color.

Starch is a component in many foods. When iodine is added to starch it turns blue. That is why, in the experiment, the solutions in both beakers turn blue. On addition of saliva which contains an amylase, a starch digesting enzyme (chemical compound that catalyses the breakdown of starch); the starch gradually converts to sugar. Once the starch is broken down, the blue colour disappears. It is important to know that the salivary amylase requires an alkaline medium to act.

Its main objective is to purify water through HRCM (A unique nano-carbon sorbent of the 21st Century) technology and to set-up water filters for the welfare of the humanity at large. During 3 days visit to Russia Mr. Gadkari was demonstrated an in-depth understanding of the Water Filtration process invented by Prof.Petrik using the High Reactive Carbon Mixture (HRCM). These demonstrations comprised of the filtration of tap water, industrial waste water, crude oil mixed with soil and even Coca Cola through HRCM filters.
