They approached his car, which had stopped on the Calder Freeway just after 11am, and found he had shot himself.
A police spokesman said the man appeared to have shot himself at some time before police approached the vehicle.
"Exactly where and when is yet to be determined," the spokesman said.
The man was arrested and has been taken to hospital in a serious condition.
It's believed he fired several shots outside the Romantics brothel.
The man fired several shots at the brothel about 9.50am. It which wasn't due to open until 10am.
The man had already fled the Colebrook Street brothel by the time police arrived and no one was injured in the shooting.
Superintendent Tony Langdon said police were still investigating the Taylors Lakes man's connection to the brothel.
He said police and special operations group officers were following the man "at a distance" on the Calder Freeway when he pulled over and emerged from his vehicle with a gunshot wound.
"At this stage it's too early to say what the reason for that is. It's very unusual circumstances."
Superintendent Langdon confirmed the man had shot at a person outside the business who then ran inside.
