Deoxyribonucleic acid (ⁱ/diˈɒksiˌraɪboʊnjʊˌkliːɪk, -ˌkleɪɪk/;DNA) is a molecule that carries most of the genetic instructions used in the development, functioning and reproduction of all known living organisms and many viruses. DNA is a nucleic acid; alongside proteins and carbohydrates, nucleic acids compose the three major macromolecules essential for all known forms of life. Most DNA molecules consist of two biopolymer strands coiled around each other to form a double helix. The two DNA strands are known as polynucleotides since they are composed of simpler units called nucleotides. Each nucleotide is composed of a nitrogen-containing nucleobase—either cytosine (C), guanine (G), adenine (A), or thymine (T)—as well as a monosaccharide sugar called deoxyribose and a phosphate group. The nucleotides are joined to one another in a chain by covalent bonds between the sugar of one nucleotide and the phosphate of the next, resulting in an alternating sugar-phosphate backbone. According to base pairing rules (A with T, and C with G), hydrogen bonds bind the nitrogenous bases of the two separate polynucleotide strands to make double-stranded DNA. The total amount of related DNA base pairs on Earth is estimated at 5.0 x 10³⁷, and weighs 50 billion tonnes. In comparison, the total mass of the biosphere has been estimated to be as much as 4 TtC (trillion tons of carbon).

Bone marrow

Bone marrow is the flexible tissue in the interior of bones. In humans, red blood cells are produced by cores of bone marrow in the heads of long bones in a process known as hematopoiesis. On average, bone marrow constitutes 4% of the total body mass of humans; in an adult having 65 kilograms of mass (143 lbs), bone marrow typically accounts for approximately 2.6 kilograms (5.7 lb). The hematopoietic component of bone marrow produces approximately 500 billion blood cells per day, which use the bone marrow vasculature as a conduit to the body's systemic circulation. Bone marrow is also a key component of the lymphatic system, producing the lymphocytes that support the body's immune system.

Bone marrow transplants can be conducted to treat severe diseases of the bone marrow, including certain forms of cancer such as leukemia. Additionally, bone marrow stem cells have been successfully transformed into functional neural cells, and can also potentially be used to treat illnesses such as inflammatory bowel disease.

First described by James Watson and Francis Crick in 1953, DNA is the famous storehouse of genetics that establishes each organism's physical characteristics. DNA (deoxyribonucleic acid) is a double-stranded molecule that is twisted into a helix structure (similar to a spiral staircase). The DNA molecule is comprised of chemical bases arranged in approximately 3 billion precise sequences. Even the DNA molecule for the single-celled bacterium, E. coli, contains enough information to fill an entire set of Encyclopedia Britannica. The four bases that make up the stairs in the spiraling staircase are adenine (A), thymine (T), cytosine (C), and guanine (G). These stairs act as the "letters" in the genetic alphabet, producing detailed instructions for the formation and functioning of the host cell. The A, T, C and G in the genetic code of the DNA molecule can be compared to the "0" and "1" in the binary code of computer software. Like software to a computer, DNA is a genetic language that communicates information to organic matter. Although DNA code is remarkably complex, the information translation system that interprets DNA is what really baffles science! Like any language, letters and words mean nothing outside the shared convention that gives those letters and words meaning. Indeed, the DNA molecule is an intricate, microscopic, digital message system with its own inherent language convention! What is a chromosome? To solve problems that explain and predict traits and variations, we first have to understand cells. All living organisms are composed of cells. Cells work like little factories doing all the jobs inside your body that are needed to keep your body functioning. Your body is made up of many different kinds of cells such as skin cells, muscle cells, and nerve cells. Some cells look like squashed bricks, some look like doughnuts, and many have irregular shapes. However, every cell, no matter what its job, has the same basic parts. All human cells have an outer border that is the boundary of the cell called the cell membrane. A liquid material called cytoplasm is inside the cell membrane, and there is a large structure suspended in the cytoplasm called the nucleus. The nucleus is the part of a cell that contains the genetic information. The nucleus is surrounded by a nuclear membrane that, like the cell membrane, makes a boundary around the nucleus. Now let's explore chromosomes. Chromosomes are thread-like structures located inside the nucleus of animal and plant cells. Each chromosome is made of protein and a single molecule of deoxyribonucleic acid, or "DNA". Passed from parents to offspring, DNA contains the specific instructions that make each type of living creature unique. The term chromosome comes from the Greek words for color (chroma) and body (soma). Scientists gave this name to chromosomes because they are cell structures, or bodies, that are strongly stained by some colorful dyes used in research. So what do chromosomes actually do? The unique structure of chromosomes keeps DNA tightly wrapped around spool-like proteins, called histones. Without such packaging, DNA molecules would be too long to fit inside cells. For an organism to grow and function properly, cells must constantly divide to produce new cells to replace old, worn-out cells. During cell division, it is essential that DNA remains intact and evenly distributed among cells. Chromosomes are a key part of the process that ensures DNA is accurately copied and distributed in the vast majority of cell divisions.

Each step is crucial to successful replication and is therefore a potential target of antiretroviral drugs. Step one is the infection of a suitable host-cell, such as a CD4-positive T-lymphocyte. Entry of HIV into the cell requires the presence of certain receptors on the cell surface, CD4 -- receptors and co-receptors such as CCR5 or CXCR4. These receptors interact with protein-complexes, which are embedded in the viral envelope. These complexes are composed of two glycoproteins: an extracellular gp 120 and a transmembrane gp 41 When HIV approaches the target cell gp120 binds to the CD4-receptors. This process is termed attachment. It promotes further binding to a co-receptor. Co-receptor binding results in a conformational change in gp120. This allows gp41 to unfold and insert its hydrophobic terminus into the cell membrane. Gp 41 then folds back on itself. This draws the virus towards the cell and facilitates the fusion of their membranes. The viral nucleocapsid enters the host cell and breaks open releasing two viral RNA-strands and 3 essential replication enzymes: Integrase, Protease and Reverse Transcriptase. Reverse Transcriptase begins the reverse transcription of viral RNA. It has two catalytic domains: The Ribonuclease-H active site And the polymerase active site Here single stranded viral RNA is transcribed into an RNA-DNA double helix. Ribonuclease- H breaks down the RNA. The polymerase then completes the remaining DNA-strand to form a DNA -- double helix. Now Integrase goes into action. It cleaves a dinucleotide from each 3-prime end of the DNA creating two sticky ends. Integrase then transfers the DNA into the cell nucleus and facilitates its integration into the host cell genome. The host cell genome now contains the genetic information of HIV. Activation of the cell induces transcription of proviral DNA into messenger RNA. The viral messenger RNA migrates into the cytoplasm where building blocks for a new virus are synthesised. Some of them have to be processed by the viral protease. Protease cleaves longer proteins into smaller core proteins. This step is crucial to create an infectious virus. Two viral RNA-strands and the replication enzymes then come together and core proteins assemble around them forming the capsid. This immature particle leaves the cell acquiring a new envelope of host and viral proteins. The virus matures and becomes ready to infect other cells. HIV replicates billions of times per day destroying the hosts` immune cells and eventually causing disease progression. Drugs which interfere with the key steps of viral replication can stop this fatal process. Entry into the host cell can be blocked by fusion inhibitors for example. Inhibition of reverse transcriptase by nucleoside inhibitors or by non-nucleoside Reverse Transcriptase- inhibitors is part of standard antiretroviral regimens. The action of Integrase can be blocked. Protease inhibitors are also part of standard antiretroviral therapy. Each blocked step in viral replication is a step towards better control of HIV disease.
