Fluorescence-activated cell sorting (FACS) is a specialized type of flow cytometry. It provides a method for sorting a heterogeneous mixture of biological cells into two or more containers, one cell at a time, based upon the specific light scattering and fluorescent characteristics of each cell. It is a useful scientific instrument as it provides fast, objective and quantitative recording of fluorescent signals from individual cells as well as physical separation of cells of particular interest. The acronym FACS is trademarked and owned by Becton, Dickinson and Company.[21] Among the large majority of researchers who use this technology for sorting or analysis, this term has become generic in common usage, much like xerox or kleenex. The first cell sorter was invented by Mack Fulwyler in 1965, using the Coulter principle, a relatively difficult technique that is no longer used in modern instruments. The technique was expanded by Len Herzenberg, who was responsible for coining the term FACS.[22] Herzenberg won the Kyoto Prize in 2006 for his seminal work in flow cytometry.
The cell suspension is entrained in the center of a narrow, rapidly flowing stream of liquid. The flow is arranged so that there is a large separation between cells relative to their diameter. A vibrating mechanism causes the stream of cells to break into individual droplets. The system is adjusted so that there is a low probability of more than one cell per droplet. Just before the stream breaks into droplets, the flow passes through a fluorescence measuring station where the fluorescent character of interest of each cell is measured. An electrical charging ring is placed just at the point where the stream breaks into droplets. A charge is placed on the ring based on the immediately prior fluorescence intensity measurement, and the opposite charge is trapped on the droplet as it breaks from the stream. The charged droplets then fall through an electrostatic deflection system that diverts droplets into containers based upon their charge. In some systems, the charge is applied directly to the stream, and the droplet breaking off retains charge of the same sign as the stream. The stream is then returned to neutral after the droplet breaks off.

The Principles of Flow Cytometry and FACS.

Flow cytometry is a technique by which light is used to count and characterize cells in a heterogeneous population. This is a powerful tool used in technology employed in cell counting, cell sorting, biomarker detection, and more.
While you may have heard term flow cytometry be used interchangeably with FACS, these are not completely the same. FACS, or fluorescence activated cell sorting is a type of flow cytometry in which the cells in the sample are physically sorted into different groups after analysis.
In order to perform flow cytometry, you must have an appropriate sample. This sample must be made of single cells, and not attached tissues so that the individual cellular analysis can be performed. Furthermore, incubation with fluorescent probes can be used to identify cells with certain markers or characteristics. For example, a particular cell surface protein can be detected.
The overall flow cytometry analyzer is made up of a fluidic system, optics, electronics, and sorting in the case of FACS.
The fluidic system introduces the cells to the process.
In order to illuminate each cell to obtain cellular information, we need to be able to illuminate each cell individually. This is managed by the fluidics system. The fluidics system is made of a central core through which sample fluid injected. This is enclosed by an outer sheath fluid. Both of these fluids are pushed through the system at slightly different pressures. Since the sheath fluid is under higher pressure, it moves faster, creating a drag effect causing the sample in the central core to narrow. This process is called hydrodynamic focusing and results in a single line of particles. Without hydrodynamic focusing, the nozzle of the instrument (typically 70 or 130 µm) would become blocked.
The optics involves the light applied to the sample as well as a combination of mirrors and detectors.
When the laser is reaches each cell (called the interrogation point) light is scattered and fluorescence is also emitted.
Forward scattering, as the name implies, refers to scattering of light in the direction of the initial light path. This is collected by a forward scatter detector and is indicative of cell size. A stronger forward scatter signal corresponds with a larger cell size. For example,
Light is not only scattered forward, but in all directions. This side scatter is picked up by a side scatter detector. Side scattering signal is used to describe the granularity or complexity of a cell. Highly granular cells with a large amount of internal complexity, like neutrophils, will produce more side-scattered light
Light can also trigger fluorescence emission if a cell is naturally fluorescent or has been tagged with fluorescent molecules.
There are a range of mirrors and detectors that collect this light, convert it to electrical pulses, and then take it to a computer to be analyzed. A system of filters ensures that each photodetector receives light bands of various wavelengths.
The electronic system converts photons to photoelectrons and converts the analog signal into a digital one.
The interaction of the laser bean with the cell results in a voltage pulse. The peak increases as the cell approaches the laser and decreases as it leaves.
Thresholding is used to neglect voltage pulses due to debris and small particulate matter. A threshold that is too low will introduce noise to the results. And a threshold that is too high will ignore cells of interest.
If using FACs, a sorting step is also involved in the process.
Cell sorting is based on parameters decided by the user. Based on the measured fluorescent intensity measured, an electrical charge will be applied to that cell. Electromagnets then guide the cells based on charge to the appropriate fraction.
Overall in this process the user is able to control cell sorting conditions as well as the markers that are fluorescently tagged
Measured intensity values will provide information on size, cell granularity, as well as the presence of fluorescence based on what is being tagged.
In summary the process starts with a single cells suspension that is passed through a narrow channel one at a time. Laser light illuminates individual cells in the channel and scattered light as well as fluorescent emission is picked up by detectors. These detectors in convert this information to a digital signal. This signal can then be analyzed by the user. Additionally, information from this signal can be combined with user defined parameters to sort cells. FACS uses electrodes to impose an electrical charge on each cell. After exiting the flow chamber. Electromagnets will sort cells based on charge.

It is a useful scientific instrument as it provides fast, objective and quantitative recording of fluorescent signals from individual cells as well as physical separation of cells of particular interest. The acronym FACS is trademarked and owned by Becton, Dickinson and Company.[21] Among the large majority of researchers who use this technology for sorting or analysis, this term has become generic in common usage, much like xerox or kleenex. The first cell sorter was invented by Mack Fulwyler in 1965, using the Coulter principle, a relatively difficult technique that is no longer used in modern instruments. The technique was expanded by Len Herzenberg, who was responsible for coining the term FACS.[22] Herzenberg won the Kyoto Prize in 2006 for his seminal work in flow cytometry.
The cell suspension is entrained in the center of a narrow, rapidly flowing stream of liquid. The flow is arranged so that there is a large separation between cells relative to their diameter. A vibrating mechanism causes the stream of cells to break into individual droplets. The system is adjusted so that there is a low probability of more than one cell per droplet. Just before the stream breaks into droplets, the flow passes through a fluorescence measuring station where the fluorescent character of interest of each cell is measured. An electrical charging ring is placed just at the point where the stream breaks into droplets. A charge is placed on the ring based on the immediately prior fluorescence intensity measurement, and the opposite charge is trapped on the droplet as it breaks from the stream. The charged droplets then fall through an electrostatic deflection system that diverts droplets into containers based upon their charge. In some systems, the charge is applied directly to the stream, and the droplet breaking off retains charge of the same sign as the stream. The stream is then returned to neutral after the droplet breaks off.

Flow cytometry is a technique by which light is used to count and characterize cells in a heterogeneous population. This is a powerful tool used in technology employed in cell counting, cell sorting, biomarker detection, and more.
While you may have heard term flow cytometry be used interchangeably with FACS, these are not completely the same. FACS, or fluorescence activated cell sorting is a type of flow cytometry in which the cells in the sample are physically sorted into different groups after analysis.
In order to perform flow cytometry, you must have an appropriate sample. This sample must be made of single cells, and not attached tissues so that the individual cellular analysis can be performed. Furthermore, incubation with fluorescent probes can be used to identify cells with certain markers or characteristics. For example, a particular cell surface protein can be detected.
The overall flow cytometry analyzer is made up of a fluidic system, optics, electronics, and sorting in the case of FACS.
The fluidic system introduces the cells to the process.
In order to illuminate each cell to obtain cellular information, we need to be able to illuminate each cell individually. This is managed by the fluidics system. The fluidics system is made of a central core through which sample fluid injected. This is enclosed by an outer sheath fluid. Both of these fluids are pushed through the system at slightly different pressures. Since the sheath fluid is under higher pressure, it moves faster, creating a drag effect causing the sample in the central core to narrow. This process is called hydrodynamic focusing and results in a single line of particles. Without hydrodynamic focusing, the nozzle of the instrument (typically 70 or 130 µm) would become blocked.
The optics involves the light applied to the sample as well as a combination of mirrors and detectors.
When the laser is reaches each cell (called the interrogation point) light is scattered and fluorescence is also emitted.
Forward scattering, as the name implies, refers to scattering of light in the direction of the initial light path. This is collected by a forward scatter detector and is indicative of cell size. A stronger forward scatter signal corresponds with a larger cell size. For example,
Light is not only scattered forward, but in all directions. This side scatter is picked up by a side scatter detector. Side scattering signal is used to describe the granularity or complexity of a cell. Highly granular cells with a large amount of internal complexity, like neutrophils, will produce more side-scattered light
Light can also trigger fluorescence emission if a cell is naturally fluorescent or has been tagged with fluorescent molecules.
There are a range of mirrors and detectors that collect this light, convert it to electrical pulses, and then take it to a computer to be analyzed. A system of filters ensures that each photodetector receives light bands of various wavelengths.
The electronic system converts photons to photoelectrons and converts the analog signal into a digital one.
The interaction of the laser bean with the cell results in a voltage pulse. The peak increases as the cell approaches the laser and decreases as it leaves.
Thresholding is used to neglect voltage pulses due to debris and small particulate matter. A threshold that is too low will introduce noise to the results. And a threshold that is too high will ignore cells of interest.
If using FACs, a sorting step is also involved in the process.
Cell sorting is based on parameters decided by the user. Based on the measured fluorescent intensity measured, an electrical charge will be applied to that cell. Electromagnets then guide the cells based on charge to the appropriate fraction.
Overall in this process the user is able to control cell sorting conditions as well as the markers that are fluorescently tagged
Measured intensity values will provide information on size, cell granularity, as well as the presence of fluorescence based on what is being tagged.

It would not be wrong to say that more than half of the internet is made of cat videos and pictures.
