Five people have been arrested following a crash that left a man with serious injuries and two separate car chases across Adelaide.

Police said a Holden Barina rolled after it crashed into the rear of a truck travelling on the Southern Expressway in Adelaide's south just before 1:00pm.

The car driver was taken to hospital with serious injuries while two passengers were treated for non-life threatening injuries.

Chief Inspector Julie Thomas said police received reports that a black Mercedes was seen on the expressway at the time.

"We had reason to be looking for that vehicle as of a result of this incident here and once we had located that vehicle a pursuit ensued," she said.

It is alleged the car was spotted about an hour later on Henley Beach Road at Thebarton with about five occupants inside.

Patrol cars and the police helicopter pursued the car across a number of suburbs before it was allegedly abandoned at Dry Creek.

A man and two women were arrested but two other men allegedly stole a Nissan SUV and escaped.

The SUV was spotted a short time later on Alderberry Avenue at Salisbury North and pursued across the suburbs before it was crashed into a ditch near the intersection of West and Purling Avenues.

Two men were arrested at the scene.

Police are investigating any links between the Southern Expressway crash and the allegedly stolen black Mercedes.

"There'll be traffic-related charges in relation to their manner of driving and there'll be criminal offences in relation to the stolen vehicles," chief inspector Thomas said.
