Police have attended a fatal car crash in rural South Australia.

A teenage boy and three men have lost their lives in a horror car crash in regional South Australia.

The group --- males aged 17, 22, 23 and 28 -- were in the car when it rolled and left the road on the Riddoch Highway near Penola, about 340 kilometres southeast of Adelaide. All those travelling in the car were killed in the smash, SA police said.

The shocking incident occurred about 3am Saturday, the Nine Network reports.

Emergency crews have closed the highway south of Penola and the road is expected to remain closed for some time.

"Through traffic especially heavy vehicles are asked to use the Penola Bypass. The main street is only accessible for local traffic," police are quoted as saying.

"Some diversions are in place, those travelling south on Riddoch Highway turn right onto Robe Road, then turn left on the Penola Bypass. Please avoid the area if you can."
