Parker was born in Fort Jackson, South Carolina. The youngest of four children, she is the daughter of Caroline Louise (née Morell) and John Morgan Parker, a judge who served in the U.S. Army. Her ancestry includes Swedish (from her maternal grandfather), English, Scottish, Scots-Irish, German, and Dutch. Because of her father's career, Parker spent parts of her childhood in Tennessee and Texas, as well as in Thailand, Germany, and France. She described her childhood as "profoundly unhappy", further noting that, "My parents did everything they could; I had books, clothes, a home and a warm bed, but I was never happy". She graduated from Marcos de Niza High School in Tempe, Arizona. Parker majored in drama at the North Carolina School of the Arts and graduated in 1986.

Ferguson has also written two books: Between the Bridge and the River, a novel, and American on Purpose, a memoir.
Ferguson was born in Stobhill Hospital, in the Springburn district of Glasgow to Robert and Janet Ferguson, on 17 May 1962, and raised in nearby Cumbernauld, growing up "chubby and bullied". He was raised Presbyterian. When he was six months old, he and his family moved from their Springburn apartment to a council house in Cumbernauld. They lived there as Glasgow was re-housing many people following damage to the city from World War II. Ferguson attended Muirfield Primary School and Cumbernauld High School. After an acclaimed off-Broadway run in 2015, Mary-Louise Parker and Denis Arndt have taken the stage of Broadway's Samuel J. Friedman Theatre in Simon Stephens' Heisenberg. Following the uncertain romance between a fortysomething woman and a much older man, the work is imbued with a great deal of hope and humanity, as the stars, their playwright, and director Mark Brokaw were eager to point out on opening night. TheaterMania caught up with them at the Copacabana shortly after the curtain came down.
