A hepatocyte is a cell of the main parenchymal tissue of the liver. Hepatocytes make up 70-85% of the liver's mass. These cells are involved in:
The typical hepatocyte is cubical with sides of 20-30 µm, (in comparison, a human hair has a diameter of 17 to 180 µm). The typical volume of a hepatocyte is 3.4 x 10−9 cm3.
Smooth endoplasmic reticulum is abundant in hepatocytes, whereas most cells in the body have only small amounts.
Hepatocytes display an eosinophilic cytoplasm, reflecting numerous mitochondria, and basophilic stippling due to large amounts of rough endoplasmic reticulum and free ribosomes. Brown lipofuscin granules are also observed (with increasing age) together with irregular unstained areas of cytoplasm; these correspond to cytoplasmic glycogen and lipid stores removed during histological preparation. The average life span of the hepatocyte is 5 months; they are able to regenerate.

The Architecture of the Liver The liver is the largest organ in the body. Liver tissue is composed of a compact mass of multisided units, known as the hepatic lobules. Each lobule consists of a central vein, which acts as a tributary of the hepatic vein and conducts processed blood away from the lobule, surrounded by plates of liver cells. The liver receives blood from two sources, 80 percent, which carries digested food materials, arrives from the intestine via the portal vein and venules; the remaining twenty percent is oxygenated blood from the heart, which enters through the hepatic artery and arterioles. This clip details the first step in the surgical procedure for isolation of primary hepatocytes, following exposure of the internal organs. The portal vein is cannulated with a 23 ga needle, buffer (HBSS) is perfused through the vein, the IVC is cut for drainage, and in subsequent steps (not shown here), the liver is digested with collagenase to liberate hepatocytes. The mouse is completely anesthetized long before the surgical procedure commences, and all steps are in compliance with IACUC protocols. This is, obviously, non-survival surgery.
