A fibroblast is a type of cell that synthesizes the extracellular matrix and collagen, the structural framework (stroma) for animal tissues, and plays a critical role in wound healing. Fibroblasts are the most common cells of connective tissue in animals.

Dermal Fibroblasts are responsible for producing the extracellular matrix forming the connective tissue of the skin, and play a crucial role during wound healing.
At Cell Applications, Inc., primary Human Dermal Fibroblasts are isolated from the dermis of normal human neonatal foreskin or adult skin*. They are cryopreserved at the end of primary culture and can be cultured and propagated for at least 16 population doublings. The cells are tested positive for fibroblast surface protein, and can be shipped frozen or proliferating, with all the media reagents included. HDF provide a well-established system for in vitro analysis of fibroblast growth, migration and collagen metabolism.(1-4) Fibroblasts grown in a biodegradable mesh have been used as a living dermal replacement.(5,6)

Suggested conclusion: under states of chronic silent inflammation or in an acidic condition (like under stress) these cells have a higher contractility, and may tend to tighten the collagen net more than during normal conditions. If so, then a few days of such cellular activity may be sufficient to induce a chronic fascial contracture which may lead to myofascial stiffness and reduced jont mobility.

Technical description: Clip shows mammary architecture and microenvironment engineering or MAME co-culture of MCF10.DCIS cells with CAF40TKi fibroblasts. Computer curser is used to outline a CAF (single cell) and the tumor spheroids (multiple identified in video). Benign tumor spheroids have a characteristic rotating motion that is noticed immediately. CAFs, only two in the field of view, tend to migrate freely in the 3D matrix. The CAFs extend fingerlike projections up to 100 microns in length. This timelapse video was captured using a Perkin Elmer UltraVIEW ERS confocal microscope.

Cancer-associated fibroblast converts benign non-invasive breast tumor cells into an invasive phenotype. These cells are being cultured in a 3-dimensional matrix and imaged in real-time.

Technical description: Clip shows mammary architecture and microenvironment engineering or MAME co-culture of MCF10.DCIS cells with CAF40TKi fibroblasts. Computer curser is used to outline a CAF (single cell) and the tumor spheroids (multiple identified in video). Benign tumor spheroids have a characteristic rotating motion that is noticed immediately. CAFs, only two in the field of view, tend to migrate freely in the 3D matrix. The CAFs extend fingerlike projections up to 100 microns in length. This timelapse video was captured using a Perkin Elmer UltraVIEW ERS confocal microscope.

Human Fibroblast-Like Synoviocytes isolated from an RA patient undergoing normal growth in culture (for 4 days post-thawing, with medium changed every other day). Note the elongated fibroblast-like shape of the cells and healthy cell divisions throughout the observation period.
Fibroblast-like synoviocytes* are found in the joints, where they are responsible for producing glycoproteins that make up most of the synovial fluid, essential for joint lubrication. Like the name suggests, these cells are related to fibroblasts and share the origins and some of the markers with them. During rheumatoid arthritis, the joint becomes constantly inflamed, and, concurrently, synoviocytes acquire abnormal features and altered gene expression, contributing to destruction of the joint tissue.

They are cryopreserved at second passage and can be cultured and propagated at least 5 population doublings. The cells can be shipped frozen or proliferating, with all the media reagents included. HFLS are long known for their role in joint destruction by producing cytokines and metalloproteinases that contribute to cartilage degradation.(1,2) Proinflammatory cytokines induce the proliferation, collagenase and aggrecanase production and GM-CSF secretion on HFLS.(3,4) Ongoing arthritis research also has shown that HFLS express apoptosis and P53 mutations.(5,6)
Cell Applications, Inc. provides three types of HFLS that are useful cellular models for studying the differences between HFLS-RA and HFLS-OA, such as the expression and regulation of proteases(7) and integrin subunits.(8).
