Overview:
Publications describing the effects
of decalcification on immunohistochemistry (
IHC) staining have been published intermittently with limited new data since the
1990s. At the same time IHC stains have become an ever more important and dependent tool in anatomic pathology for diagnosis, prognosis, and treatment purposes.
Current instrumentation, in addition to being fully automated, typically use polymer based detection systems, in place of avidin-biotin complex based detection, which was predominate when most of the previous studies were performed. This presentation will review performance of
ISH and IHC markers decalcification reagents, and antibodies with current detection system of polymer based detection. Also, a review of current IHC validation recommendation concerning decalcified tissue will be discussed.
Learning Objectives:
Characterize performance of ISH markers; kappa and lambda, with current decalcification reagents will be reviewed.
Characterize performance of 33 different IHC markers with current decalcification reagents will be reviewed.
Review current IHC validation recommendations concerning decalcified tissue currently in the
USA.
Biography:
Philip Ferguson,
M.D. is a practicing pathologist in private practice located in
Arkansas. He has a diverse training background that includes an undergraduate degree in mechanical engineering, post-graduate training in internal medicine, and board certifications in anatomic pathology, clinical pathology, and hematopathology.
His professional interests center around educational resource development for pathologists and trainees, and use of immunohisochemistry and molecular diagnostics in surgical pathology. Dr. Ferguson launched one of the first widespread internet educational tools for pathology residents to prepare for their board examination, PathMD (www.PathMD.com), which is now the Leica tool PathLead. Dr. Ferguson also serves as a state commissioner for the
College of American Pathologists' Accreditation and Laboratory Improvement
Program and is also member of the
Global Advisory Board for
Leica Biosystems. Dr. Ferguson has published several publication of late including ‘
The Effect of
Decalcification Solutions on
Kappa and
Lambda In-Situ
Hybridization,’ as well as ‘
Effects of Decalcification on Immunohistochemistry Comparing: Immunocal®, Formical2000®, and
EDTA Stat®.’
