Rapid Screening of Gene Function by Systemic Delivery of Morpholino Oligonucleotides to Live Mouse Embryos. Kathryn S. McClelland et al (2015), PLoS ONE, http://dx.doi.org/10.1371/journal.pone.0114932 Traditional gene targeting methods in mice are complex and time consuming, especially when conditional deletion methods are required. Here, we describe a novel technique for assessing gene function by injection of modified antisense morpholino oligonucleotides (MOs) into the heart of mid-gestation mouse embryos. After allowing MOs to circulate through the embryonic vasculature, target tissues were explanted, cultured and analysed for expression of key markers. We established proof-of-principle by partially phenocopying known gene knockout phenotypes in the fetal gonads (Stra8, Sox9) and pancreas (Sox9). We also generated a novel double knockdown of Gli1 and Gli2, revealing defects in Leydig cell differentiation in the fetal testis. Finally, we gained insight into the roles of Adamts19 and Ctrb1, genes of unknown function in sex determination and gonadal development. These studies reveal the utility of this method as a means of first-pass analysis of gene function during organogenesis before committing to detailed genetic analysis.

A new compound developed by researchers from the University of Iowa and the Iowa City VA Medical Center has been shown to help muscles burn calories more effectively by suppressing their innate impulse to store more energy. An injection has been shown to override the body’s natural impulse to retain energy in its muscles, providing a possible new tool to jumpstart weight loss. The research, conducted by a team from the University of Iowa and the Veterans Affairs Medical Center in Iowa City, aims to counteract the body’s often frustrating reaction to a reduction in calories which is to improve its functional efficiency and retain as much weight as possible. Scientists targeted the protein KATP channels, which have been identified as key in maintaining muscular energy in previous research. They developed a substance called vivo-morpholino to target genes and suppress the production of KATP at a specific site only. The compound was injected into one hind leg of test mice, and the muscles in that area showed an increase in energy under infrared imaging compared to untreated mice while doing mild exercise. Though scientists think this injection could possibly help people burn more calories during daily activities, they caution that it should not be considered a substitute for exercise.

Morpholinos block small (~25 base) regions of the base-pairing surfaces of ribonucleic acid (RNA). Most Morpholinos are used as research tools for reverse genetics by knocking down gene function.
This article discusses only the Morpholino antisense oligos, which are nucleic acid analogs. The word "morpholino" can occur in other chemical names, referring to chemicals containing a six-membered morpholine ring. To help avoid confusion with other morpholine-containing molecules, when describing oligos "Morpholino" is often capitalized as a trade name, though this usage is not consistent throughout the scientific literature. Morpholinos are sometimes referred to as "PMO" (phosphorodiamidate morpholino oligo).
