Melanocyte protein PMEL also known as premelanosome protein (PMEL) or silver locus protein homolog (SILV) is a protein that in humans is encoded by the PMEL gene. Its gene product may be referred to as PMEL, silver, ME20, gp100 or Pmel17.
PMEL is a 100 kDa type I transmembrane glycoprotein that is expressed primarily in pigment cells of the skin and eye. The transmembrane form of PMEL is modified in the secretory pathway by elaboration of N-linked oligosaccharides and addition and modification of O-linked oligosaccharides. It is then targeted to precursors of the pigment organelle, the melanosome, where it is proteolytically processed to several small fragments. Some of these fragments form non-pathological amyloid that assemble into sheets and form the striated pattern that underlies melanosomal ultrastructure. PMEL cleavage is mediated by several proteases including a proprotein convertase of the furin family, a "sheddase" that might include members of the a disintegrin and metalloproteinase (ADAM) family, and additional proteases in melanosomes or their precursors. After the amyloidogenic region is cleaved, the small remaining integral membrane fragment is digested by γ-secretase.

My range review of the Ruger GP-100 revolver in .357 Magnum. This is the stainless 5" barrel model that is a distributor exclusive for Davidson's. The Standard GP-100 comes in 3, 4 and 6 inch lengths. I chose the 5' because it is a nice mix of portability and barrel length for the .357 magnum cartridge. It also has and excellent balance. The 5' 357 was a favorite of past gun writers and pistoleers like Skeeter Skelton and Elmer Keith. The Gp100 is a perfect frame size for shooting a steady diet of full power .357 loads without the risk of forcing cone damage which is common in K frame sized guns like the Smith and Wesson Model 13 M&P; and 19 Combat Masterpiece and the Ruger Security/ Service/ Speed Six. Me and my favorite sidearm, the Ruger GP100. It may not be as blue collar as most of what I show here, but for someone (non enthusiast) who may be interested in owning only one or two guns, it is a great option worth considering. If you ever have the privilege to shoot a GP100, you will not be disappointed.
Review (including some shooting) of the Ruger GP100 .357 Magnum revolver, 3" barrel. This is model #1715, satin stainless with the Hogue Monogrip. Shooting portion is up to 40 yards, using 158 grain jacketed soft point ammo. This is a very well-built revolver and does a good job of minimizing recoil. Although my primary utility will be using the GP100 for ammo testing, it would make an excellent home-defense choice and could be considered for concealed-carry.
