Each concentration used in the tests went into two replicate petri dishes. The viable embryos were then placed into petri dishes, (20 per dish) and exposed to varying concentrations of DBP dissolved in DiMethylSulfoxide (DMSO). Since the solubility of DBP is very low in water, DMSO was used as a carrier solvent to prevent the DBP from falling out of solution and thus preventing the embryos from proper exposure. Exposure times began with gastrulation and ended with free living tadpoles (96 Hrs). At the end of the assay survival, malformations and growth were recorded. Systat was used to calculate the 96 hr LC50 and EC50 using probit analysis. ANOVA was used to determine Lowest Observable Effect levels for mortality, malformation and growth. The median lethal concentration of DBP for 50% of the embryos tested in these experiments was 12.2 ppm (parts per million), while the median effective concentration required to induce malformation in 50% of the embryos was 7.8 ppm. These results confirm that DBP is a teratogen and that the FETAX assay is useful in assessing the teratogenic potential of phthalate compounds.
