We show how to extract piperine starting with black (or white) pepper by performing a microwave extraction. We obtain a crude product (3.6% of the pepper weight) and then prove it is possible to go a step further and actually isolate piperidine from this.
We perform an alkaline hydrolysis of piperine extracted from white pepper, to form piperinic acid and piperidine. We work up the reaction mixture and separate and purify both of the individual hydrolysis products.

Here’s The Missing Component That Makes Turmeric Supplementation 2000% More Bioavailable
Adding a substance found in black pepper called piperine can dramatically enhance the bioavailability of curcumin. In a US NIH study here’s what they reported:
“One of them is piperine, a major component of black pepper, known as inhibitor of hepatic and intestinal glucuronidation and is also shown to increase the bioavailability of curcumin. This effect of piperine on the pharmacokinetics of curcumin has been shown to be much greater in humans than in rats. In humans, curcumin bioavailability was increased by 2,000% at 45 minutes after co-administering curcumin orally with piperine, whereas in rats, it has been found that concomitant administration of piperine 20 mg/kg with curcumin 2 g/kg increased the serum concentration of curcumin by 154% for a short period of 1-2 hours post drug. The study shows that in the dosages used, piperine enhances the serum concentration, extent of absorption and bioavailability of curcumin in both rats and humans with no adverse effects.
Another study also showed that piperine (20 mg/kg orally) when administered with curcumin (2 g/kg orally) enhances the bioavailability of the latter up to 20-fold more in epileptic rats. Enhanced bioavailability of curcumin was also evidenced by other researcher when curcumin was administered orally concomitant with piperine. Intestinal absorption of curcumin was also found relatively higher when administered concomitantly with piperine, and it stayed significantly longer in the body tissues. In view of these findings, curcumin-piperine (Cu-Pi) nanoparticles has been prepared by various methods. The bioavailability, cellular uptake and biological effects of this nanoparticles are being tested.”

Piperine
Piperine, along with its isomer chavicine, is the alkaloid responsible for the pungency of black pepper and long pepper. It has also been used in some forms of traditional medicine and as an insecticide.
Preparation
Piperine is commercially available. If desired, it may be extracted from black pepper using dichloromethane. Aqueous hydrotopes can be used in the extraction to result in high yield and selectivity. The amount of piperine varies from 1-2% in long pepper, to 5-10% in commercial white and black peppers. Further, it may be prepared by treating the solvent-free residue from an alcoholic extract of black pepper, with a solution of potassium hydroxide to remove resin (said to contain chavicine, an isomer of piperine) and solution of the washed, insoluble residue in warm alcohol, from which the alkaloid crystallises on cooling.
Reactions
Piperine yields salts only with strong acids. The platinichloride B4•H2PtCl6 forms orange-red needles. ("B" denotes one mole of the alkaloid base in this and the following formulae.) Iodine in potassium iodide added to an alcoholic solution of the base in the presence of a little hydrochloric acid gives a characteristic periodide, B2•HI•I2, crystallising in steel-blue needles, mp. 145 °C.
