Argonaute
The Argonaute protein family plays a central role in RNA silencing processes, as essential catalytic components of the RNA-induced silencing complex (RISC). RISC complex is responsible for the gene silencing phenomenon known as RNA interference (RNAi). Argonaute proteins bind different classes of small non-coding RNAs, including microRNAs (miRNAs), small interfering RNAs (siRNAs) and Piwi-interacting RNAs (piRNAs). Small RNAs guide Argonaute proteins to their specific targets through sequence complementarity (base pairing), which then leads to mRNA cleavage or translation inhibition.
Discovery
The RNA interference (RNAi) was first reported in 1995 by Guo and Kemphues, and similar pathways collectively referred to as RNA silencing were discovered in plants and fungi. The beginning of people’s understanding of the mechanism of RNA silencing began only in 1998 with the experiments of Fire and colleagues demonstrating that double-stranded RNA triggered RNAi. RNA silencing pathways process long RNAs into small RNAs that direct the repression of transcription or translation of nucleic acid targets with sequence corresponding to the small RNAs. These single-stranded RNAs, referred to as guide strands, are incorporated into RNA silencing effectors complexes such as the RNA-induced silencing complex (RISC). These RNA silencing effector complexes contain Argonaute family proteins.

"Shock!" is the eleventh major single by Japanese idol pop group Cute. It was released on January 6, 2010, in both normal and limited editions, the limited edition containing a DVD with a version of the "Shock!" PV on it and coming with a different cover. The first press of each edition also contained a card with a serial number on it, used in an event draw to promote the single's release. This was the first single to be released after Erika Umeda's graduation, which also makes it the first single not to feature her. Airi Suzuki is "centred" in this single, taking on the main vocals. The single peaked at #1 on the Oricon daily charts, and #5 on the weekly charts. The single also reached #13 on the monthly chart for January, with a reported total of 23,389 copies sold.

A mechanical or physical shock is a sudden acceleration caused, for example, by impact, drop, kick, earthquake, or explosion. Shock is a transient physical excitation.
Shock describes matter subject to extreme rates of force wtr to time. Shock is a vector that has units of an acceleration (rate of change of velocity). The unit g (or g) represents multiples of the acceleration of gravity and is conventionally used.

Shock, also called circulatory shock, is a life-threatening clinical state characterized by body-wide deficiency of blood supply, causing oxygen deprivation, buildup of waste products, and eventual organ failure if untreated.
Shock may have different causes and hence its classification into different types:
- Hypovolemic, or low volume shock happens when the circulating blood volume is severely reduced. This can be caused by:
+ External blood loss, such as after an injury,
+ Internal blood loss such as that results from a ruptured blood vessel, ruptured ectopic pregnancy, pancreatitis …
+ Or fluid loss from major burns, excessive vomiting, diarrhea or urination.
- Cardiogenic shock occurs when the heart fails to pump sufficiently. This can result from a sudden heart attack, or an end-stage development in various heart conditions.
- Obstructive shock is caused by an obstruction of blood flow in a major circulatory circuit.
- Distributive shock results from excessive dilation of blood vessels, or vasodilation, which decreases blood pressures. Distribution shock can have different causes, the most common being sepsis, anaphylaxis and damage to the central nervous system (neurogenic):
+ In sepsis, the immune system is overwhelmed by an infection that gets out of control, and responds with a systemic cytokine release that causes vasodilation and fluid leakage from capillaries.
+ In anaphylaxis, the immune system overreacts to an allergen, releasing massive amounts of histamine, which has similar effects to cytokines. Peanut allergy is a common cause of anaphylaxis.
+ Neurogenic shock typically occurs as a result of a spinal cord injury. As the autonomic nervous system is damaged, the sympathetic tone that normally keeps blood vessels constricted is lost, causing vasodilation and hypotension.
Common symptoms of shock include low blood pressures and signs of organ damage such as confusion, reduced urine output and cold, sweaty, mottled or bluish skin, although distributive shocks due to sepsis or anaphylaxis may initially produce warm or flushed skin. This is because the infection in sepsis usually comes with fever, and the allergic reaction in anaphylaxis is accompanied by hives. Distributive shocks may also differ from other types of shock by having, at least initially, normal or high cardiac output.
As the body tries to compensate for hypotension, fast heart rates and rapid breathing may be observed. Diagnosis may also be assisted by blood tests for blood lactate levels. This is because in the absence of oxygen, the body switches to an alternative way of producing cellular energy, called anaerobic metabolism, in which glucose is broken down only partially producing lactic acid. Blood tests may also indicate signs of organ damage, or infection in case of sepsis.
Shock is a medical emergency and requires immediate treatments which aim to increase blood pressures and treat the underlying cause.

Sepsis is a life-threatening condition that occurs when the body’s excessive response to an infection causes damage to its own tissues. Sepsis may progress to septic shock, a body-wide deficiency of blood supply that leads to oxygen deprivation, buildup of waste products, and eventual organ failure. Without timely treatment, mortality rates are high.
With sepsis, patients typically experience fever, weakness, sweating, and a rapid heart rate and breathing rate. As septic shock develops, blood pressure decreases, and signs of organ damage, such as confusion and reduced urine output, can be observed. The skin is initially warm or flushed, then becomes cold, sweaty, mottled or bluish.
While any infection can lead to sepsis, bacterial infections in the lungs, digestive and urinary organs, are the most common causes. Sepsis may also develop from a post-surgery infection or an infected catheter.
Septic shock occurs more often in newborns, the elderly and pregnant women. Other risk factors include having a compromised immune system or chronic diseases, extended hospital stays, having invasive devices, and overuse of antibiotics or corticosteroids.
The pathogenesis of septic shock is not fully understood. In most cases, the immune system is overwhelmed by an infection that gets out of control, and responds with a systemic cytokine release that causes widespread vasodilation and fluid leakage from capillaries. These cytokines also activate the coagulation process, producing tiny blood clots that clog blood vessels, reducing blood flow. Bleeding may also develop because excessive coagulation depletes clotting factors. Poor capillary flow reduces oxygen supply and impairs removal of carbon dioxide and waste products, resulting in organ dysfunction and eventually failure.
Diagnosis is primarily clinical but requires confirmation of an ongoing infection.
An elevated blood lactate level serves as an indicator of shock. This is because in the absence of oxygen, the body switches to anaerobic metabolism, which breaks down glucose only partially, producing lactic acid. Blood tests may also indicate signs of organ damage, and infection. Other specimens such as urine, respiratory or wound secretions, may be taken for culture to detect infection. Imaging tests may also help identify the source of infection. Other causes of shock should be ruled out.
Early and aggressive treatment is critical for survival. Treatments include:
- Intravenous fluids, and possibly vasopressors, to restore blood flow.
- Broad-spectrum antibiotics while waiting for culture results. Once the causative organism is identified, more specific antibiotics will be used.
- Other measures to control infection.
- Supportive care such as supplemental oxygen, and in case of organ failure, mechanical ventilation or dialysis.

Shock, also called circulatory shock, is a life-threatening clinical state characterized by body-wide deficiency of blood supply, causing oxygen deprivation, buildup of waste products, and eventual organ failure if untreated.
Shock may have different causes and hence its classification into different types:
- Hypovolemic, or low volume shock happens when the circulating blood volume is severely reduced. This can be caused by:
+ External blood loss, such as after an injury,
+ Internal blood loss such as that results from a ruptured blood vessel, ruptured ectopic pregnancy, pancreatitis …
+ Or fluid loss from major burns, excessive vomiting, diarrhea or urination.
- Cardiogenic shock occurs when the heart fails to pump sufficiently. This can result from a sudden heart attack, or an end-stage development in various heart conditions.
- Obstructive shock is caused by an obstruction of blood flow in a major circulatory circuit.
- Distributive shock results from excessive dilation of blood vessels, or vasodilation, which decreases blood pressures. Distribution shock can have different causes, the most common being sepsis, anaphylaxis and damage to the central nervous system (neurogenic):
+ In sepsis, the immune system is overwhelmed by an infection that gets out of control, and responds with a systemic cytokine release that causes vasodilation and fluid leakage from capillaries.
+ In anaphylaxis, the immune system overreacts to an allergen, releasing massive amounts of histamine, which has similar effects to cytokines. Peanut allergy is a common cause of anaphylaxis.
+ Neurogenic shock typically occurs as a result of a spinal cord injury. As the autonomic nervous system is damaged, the sympathetic tone that normally keeps blood vessels constricted is lost, causing vasodilation and hypotension.
Common symptoms of shock include low blood pressures and signs of organ damage such as confusion, reduced urine output and cold, sweaty, mottled or bluish skin, although distributive shocks due to sepsis or anaphylaxis may initially produce warm or flushed skin. This is because the infection in sepsis usually comes with fever, and the allergic reaction in anaphylaxis is accompanied by hives. Distributive shocks may also differ from other types of shock by having, at least initially, normal or high cardiac output.
As the body tries to compensate for hypotension, fast heart rates and rapid breathing may be observed. Diagnosis may also be assisted by blood tests for blood lactate levels. This is because in the absence of oxygen, the body switches to an alternative way of producing cellular energy, called anaerobic metabolism, in which glucose is broken down only partially producing lactic acid. Blood tests may also indicate signs of organ damage, or infection in case of sepsis.
Shock is a medical emergency and requires immediate treatments which aim to increase blood pressures and treat the underlying cause.

Sepsis is a life-threatening condition that occurs when the body’s excessive response to an infection causes damage to its own tissues. Sepsis may progress to septic shock, a body-wide deficiency of blood supply that leads to oxygen deprivation, buildup of waste products, and eventual organ failure. Without timely treatment, mortality rates are high.
With sepsis, patients typically experience fever, weakness, sweating, and a rapid heart rate and breathing rate. As septic shock develops, blood pressure decreases, and signs of organ damage, such as confusion and reduced urine output, can be observed. The skin is initially warm or flushed, then becomes cold, sweaty, mottled or bluish.
While any infection can lead to sepsis, bacterial infections in the lungs, digestive and urinary organs, are the most common causes. Sepsis may also develop from a post-surgery infection or an infected catheter.
Septic shock occurs more often in newborns, the elderly and pregnant women. Other risk factors include having a compromised immune system or chronic diseases, extended hospital stays, having invasive devices, and overuse of antibiotics or corticosteroids.
The pathogenesis of septic shock is not fully understood. In most cases, the immune system is overwhelmed by an infection that gets out of control, and responds with a systemic cytokine release that causes widespread vasodilation and fluid leakage from capillaries. These cytokines also activate the coagulation process, producing tiny blood clots that clog blood vessels, reducing blood flow. Bleeding may also develop because excessive coagulation depletes clotting factors. Poor capillary flow reduces oxygen supply and impairs removal of carbon dioxide and waste products, resulting in organ dysfunction and eventually failure.
Diagnosis is primarily clinical but requires confirmation of an ongoing infection.
An elevated blood lactate level serves as an indicator of shock. This is because in the absence of oxygen, the body switches to anaerobic metabolism, which breaks down glucose only partially, producing lactic acid. Blood tests may also indicate signs of organ damage, and infection. Other specimens such as urine, respiratory or wound secretions, may be taken for culture to detect infection. Imaging tests may also help identify the source of infection. Other causes of shock should be ruled out.
Early and aggressive treatment is critical for survival. Treatments include:
- Intravenous fluids, and possibly vasopressors, to restore blood flow.
- Broad-spectrum antibiotics while waiting for culture results. Once the causative organism is identified, more specific antibiotics will be used.
- Other measures to control infection.
- Supportive care such as supplemental oxygen, and in case of organ failure, mechanical ventilation or dialysis.
