Contractile actomyosin bundles, stress fibers, contribute to morphogenesis, migration, and mechanosensing of non-muscle cells. In addition to actin and non-muscle myosin II (NMII), stress fibers contain a large array of proteins that control their assembly, turnover, and contractility. Calponin-3 (Cnn3) is an actin-binding protein that associates with stress fibers. Calponin-3 is critical for coordinated contractility of actin stress fibers. Katarzyna Ciuba et al (2018), Scientific Reports https://doi.org/10.1038/s41598-018-35948-6
Contractile actomyosin bundles, stress fibers, contribute to morphogenesis, migration, and mechanosensing of non-muscle cells. In addition to actin and non-muscle myosin II (NMII), stress fibers contain a large array of proteins that control their assembly, turnover, and contractility. Calponin-3 (Cnn3) is an actin-binding protein that associates with stress fibers. However, whether Cnn3 promotes stress fiber assembly, or serves as either a positive or negative regulator of their contractility has remained obscure.
Here, we applied U2OS osteosarcoma cells as a model system to study the function of Cnn3. We show that Cnn3 localizes to both NMII-containing contractile ventral stress fibers and transverse arcs, as well as to non-contractile dorsal stress fibers that do not contain NMII. Fluorescence-recovery-after-photobleaching experiments revealed that Cnn3 is a dynamic component of stress fibers. Importantly, CRISPR/Cas9 knockout and RNAi knockdown studies demonstrated that Cnn3 is not essential for stress fiber assembly.
However, Cnn3 depletion resulted in increased and uncoordinated contractility of stress fibers that often led to breakage of individual actomyosin bundles within the stress fiber network. Collectively these results provide evidence that Cnn3 is dispensable for the assembly of actomyosin bundles, but that it is required for controlling proper contractility of the stress fiber network.

How Sinn Fein’s Rise Is Shaking Up Ireland’s Politics
Rooted in its campaign for a united Ireland, Sinn Fein was long an outsider in politics due to its links with the Irish Republican Army. With the conflict in Northern Ireland largely over since a 1998 peace deal, the movement has reinvented itself to appeal to a new generation of voters.
Now, in a historic shift, it has become the biggest party in the north and also leads opinion polls in the Irish Republic to the south. That makes its demand for a referendum on unification harder to ignore.
The party, whose name means “Ourselves Alone,” was created amid the campaign for Irish home rule at the start of the 20th century. After the Catholic south won its independence from Britain in 1921, Sinn Fein split, ultimately creating what are now the three largest parties in the Republic.

Sinn Fein's Michelle O'Neill becomes Northern Ireland’s first nationalist first minister
Stormont has finally been restored and for the first time in 103 years it's not a unionist at the helm. Making history Sinn Fein's Michelle O'Neill, who's become the first nationalist to lead Northern Ireland - a state she wants to abolish. She's pledged to be a "first minister for all" and will share power with Emma Little-Pengelly from the Democratic Unionist Party - who's been nominated deputy first minister.
But two years of political paralysis, with the government effectively on auto-pilot, have left them with a long list of problems to tackle.

2023 marks the 25th anniversary of signing of The Belfast (Good Friday) Agreement, which brought an end to 30 years of conflict in Northern Ireland known as 'the Troubles'. This conflict had a deep and embedded history that stretches back over a century.
Between 1919 and 1921, Ireland played host to a long and bloody guerrilla conflict between British state forces and Irish republican guerrillas, in the form of Irish Volunteers or the Irish Republican Army (IRA). The conflict would completely transform the political landscape in Ireland.

The modern-day Sinn Fein started to seriously contest elections south of the border in the 1980s under a strategy known as the “Armalite and the Ballot Box.” (Armalite is a gun manufacturer whose weapons were used by Republican paramilitaries against British security forces).
Today, it’s a broadly center-left party that campaigns for higher government spending, better housing and increased taxes on the rich.

Rooted in its campaign for a united Ireland, Sinn Fein was long an outsider in politics due to its links with the Irish Republican Army. With the conflict in Northern Ireland largely over since a 1998 peace deal, the movement has reinvented itself to appeal to a new generation of voters.
Now, in a historic shift, it has become the biggest party in the north and also leads opinion polls in the Irish Republic to the south. That makes its demand for a referendum on unification harder to ignore.
