Taliban fighters tried to ambush a joint US-Afghan military convoy in Helmand province's Gereshk district late on Friday before fleeing into a nearby village for cover, said Mohammad Hussein, the provincial police chief.
Airstrikes targeted the militants in the village of Hyderabad, said Dur Ali Shah, the mayor of Gereshk.
Shah said 30 to 35 people were killed or wounded but he could not provide an exact breakdown.
Villagers reported far more than 30 casualties but those figures were not immediately corroborated by officials.
"Last night NATO forces conducted an airstrike on our village. More than 60 civilians from our village were killed. Seven of my family members were killed in this bombing," said Mohammad Khan, a resident of Hyderabad.
Khan said seven members of his family, including his brother and five of his brother's children, were killed by airstrikes.
"Police and Taliban fighters were fighting, there was an airstrike on the village, some members of my family were killed and others were wounded in the airstrike,'' said Divi Zohra, who was injured in the strike.
Those injured in the airstrike were taken to hospitals in the Gereshk and Lashkar Gha.
Six houses were destroyed during the clash, Shah said.
Shah said he did not know the number of Taliban casualties.
The US-led coalition acknowledged that some civilians were killed during the clash, which followed machine gun, mortar and rocker propelled grenade attacks on a joint Afghan-coalition patrol.
Civilians deaths caused by US and NATO-led troops have infuriated Afghans and prompted President Hamid Karzai to publicly condemn foreign forces for carelessness and viewing Afghan lives as cheap.
He has urged restraint and better coordination of military operations with the Afghan government, while also blaming Taliban for using civilians as human shields.
Violence has soared in Afghanistan with more than 2,800 people, mostly militants, killed in fighting this year, according to an Associated Press tally of figures from Western military and Afghan officials.
A count by the United Nations and an umbrella organisation of Afghan and international aid groups shows the number of civilians killed by international forces was slightly greater than the number killed by insurgents in the first half of the year.
An AP count based on figures from Afghan and international officials found that militants killed 178 civilians in attacks through June 23 and that Western forces killed 203.
The US and NATO say they don't have civilian casualty figures.

HYDERABAD : A former Google employee was detained for allegedly trying to join the Islamic State (IS) in Iraq, police said, after making a breakthrough which could help them bust an organised network of home-grown jihadis, planning to join the militant group.
Police sources in Hyderabad said that security analyst Munawad Salman, 30, a resident of Musheerabad, was held after officials tracked his conversation for months and was certain that his hidden agenda to visit Saudi Arabia was to cross over to Iraq and join Islamic State .
"He is being grilled for more information. This is perhaps the first time, a software engineer has got attracted to a global terror outfit like Islamic State in the recent past," a senior police official told.
"The arrest is very significant and could open a big door for us to trace more educated youth getting influenced by militants," the officer said, referring to IS, whose main motive is to establish Caliphate (Islamic State) in vast swathes of the world.
Software engineer Salman, quit his Google India job six months ago. Since then, he had been actively reading 'Al Isabha' web propaganda, which is again run by sympathizers of Islamic State , formerly Islamic State of Iraq and Syria and al-Qaida, to target Indian Muslims, sources said.
The techie is learnt to have been in touch with handlers from ISIS for nearly a year and has allegedly exchanged emails before being completely influenced by people behind 'Al Isabha'.
With security agencies on high alert across the country, especially on the look out for prospective people or sympathizers of Islamic State and al-Qaida, planning to join the outfits, they stumbled upon Salman's conversation with his handlers.
During interrogation, Salman said he had a visa to travel to Saudi Arabia and join a local company there.
"He has a proper job visa to go to Saudi Arabia, but there was clear indications that he was planning to join IS either immediately or later. The family members also counselled him to desist from getting attracted to such ideas, when they came to know about the matter," another official said.

U.S. Central Command said in a statement on Friday that an ISIS chemical weapons expert was killed in an airstrike on Saturday near Mosul, Iraq. Central Command said Abu Malik worked in a chemical weapons production facility under Saddam Hussein before joining al Qaeda in Iraq in 2005. He then joined ISIS, allowing the terrorist group to make use of his experience in chemical weapons capabilities. His death is expected to temporarily degrade and disrupt the terrorist network and diminish ISIS's ability to potentially produce and use chemical weapons against innocent people.

U.S. Central Command said in a statement on Friday that an ISIS chemical weapons expert was killed in an airstrike on Saturday near Mosul, Iraq. Central Command said Abu Malik worked in a chemical weapons production facility under Saddam Hussein before joining al Qaeda in Iraq in 2005. He then joined ISIS, allowing the terrorist group to make use of his experience in chemical weapons capabilities. His death is expected to temporarily degrade and disrupt the terrorist network and diminish ISIS's ability to potentially produce and use chemical weapons against innocent people.

Saudi-led airstrikes have driven back rebels in the last stronghold of Yemen's president, as al-Qaeda fighters seized a major army base in southeast of the country.
The setback for the Shia fighers on Friday came as warplanes from the Saudi-led coalition dropped weapons and medical aid to fighters defending the southern Yemeni city, a last symbolic foothold of President Abd-Rabbu Mansour Hadi, who fled to Saudi Arabia after the Houthis seized the capital, Sanaa.
The Houthi fighters and their allies withdrew from Crater neighbourhood, as well as one of Aden's presidential residences which they had seized a day earlier, residents and a local official said.
Their withdrawal followed overnight clashes and an airstrike on the presidential palace at Ma'ashiq, overlooking Crater. At least one Houthi tank was destroyed and another taken over by Hadi's loyalists, they said.
Around 12 Houthi fighters were killed in the clashes in Aden, but there has been no word on casualties suffered by forces loyal to Hadi.
In fighting on Friday, al-Qaeda in the Arabian Peninsula, as the Yemen branch is named, consolidated its hold of Mukalla, which they overran on Thursday, seizing its port and a major army base there.
The group has benefited from Yemen's political crisis ever since the Houthis first surged from their northern strongholds last year to seize Sanaa, and much of the north.
Brigadier General Ahmed al-Asiri, a spokesman for the Arab coalition told a news conference airstrikes had significantly degraded the Houthi fighters' military capabilities and Hadi loyalists had gained ground in Aden.
The Houthis are backed in their push by military forces and police loyal to former president Ali Abdullah Saleh.
Despite inflicting heavy losses on the Houthis, the airstrikes might not be enough to halt their advance, said Al Jazeera's Mohamed Vall, who has reported extensively on Yemen.
"Airstrikes have never really been so decisive. The Houthis can easily blend with the local population and they have almost the entire support of the Yemeni army," he said.
"They [Saudi-led coalition] know they cannot win this war without putting troops on the ground...they are probably waiting for more weakening of Houthi defences [before they do that]."
Early on Friday, warplanes from the coalition dropped crates of weapons and medical supplies by parachute over Tawahi, a district on the far end of the Aden peninsula which is still held by Hadi loyalists, fighters told the Reuters news agency.
The crates included light weapons, telecommunications equipment and rocket-propelled grenades, they said.
The coalition, which is trying to reassert Hadi's authority as a prelude to political negotiations, has repeatedly said that sending ground troops into Yemen remains an option but not an automatic move.
Officials have declined to say whether special forces have already deployed. The Saudi ambassador to Washington, Adel al-Jubeir, said on Thursday the kingdom does not have "formal" troops on the ground in Aden.
Tribal sources in Yemen said on Friday that Saudi Arabia had started to remove parts of a fence along its border with the northwestern Yemeni provinces of Saada and Hajja.
This could be a prelude to an incursion by ground troops, but may also be part of more modest efforts to secure the frontier area, which on some stretches includes a buffer zone between the two countries.
The Saudi-led coalition involves six nations that belong to the Gulf Cooperation Council. The GCC members include Saudi Arabia itself, Bahrain, Kuwaiti, Oman, Qatar and the United Arab Emirates.
