This was created over a couple of days to demonstrate how a simple animation can be used as a learning aid for scientific concepts that may be difficult for a student to grasp. The audio came out quieter than expected, so you may have to turn up your speakers slightly. In the above animation, the normal enzyme catalytic cycle is shown and is followed by two types of inhibition by competitive and non-competitive blockers. This animation shows how Prozac® alleviates depression. It can also be used to illustrate in general how neuron cells communicate with each other and how a neurotransmitter sends a signal from one neuron to another. Some people with depression have a shortage of serotonin, the "mood" neurotransmitter in the brain. The antidepressant Prozac®, a Selective Serotonin Reuptake Inhibitor (SSRI), can help correct this imbalance by increasing the brain's own supply of serotonin. This animation shows how Prozac® acts as a selective inhibitor of Serotonin Reuptake Transporter Protein, thus alleviating depression. In the brain, serotonin is associated with transmission of thoughts and feelings. In a healthy person, an optimal concentration of serotonin is available at the synapse. The imbalance of this neurotransmitter triggers emotional symptoms, like depressed mood, or physical symptoms, like aches and pains. The blue colored layers represent the trans-membrane structure of both pre- and post-synaptic areas (the upper and lower part of the screen, respectively). Red colored masses in the post-synaptic membrane represent serotonin receptors. There are other membrane proteins as well. Depression can occur when the serotonin transporter protein (a G-protein coupled receptor; shown in white in the pre-synaptic membrane) takes up a serotonin molecule before it has a chance to bind to the post-synaptic receptor. PARP inhibitors
Prof Helleday and Prof Chalmers speak to ecancer.tv about PARP inhibitors, their significance to cancer treatment and their potential for use in combination with radiotherapy. PARP inhibitors allow clinicians to target treatment specifically at the cancer cells which dramatically reduces side effects. Madrid, Spain: Compounds in marijuana inhibit malignant brain tumor growth in animals, and may provide a potential therapy for human glioma patients, according to a clinical review appearing in the September issue of Neuropharmacology. "Current therapeutic strategies for the treatment of [gliomas] are usually ineffective or just palliative," researchers from the Department of Biochemistry and Molecular Biology at Madrid's Complutense University wrote. "During the last few years, several studies have shown that cannabinoids ... slow the growth of different types of tumours, including gliomas, in laboratory animals. Cannabinoids induce apoptosis (programmed cell death) of glioma cells [in vitro.] In addition, cannabinoid treatment inhibits angiogenesis (growth) of gliomas in vivo. Remarkably, cannabinoids kill glioma cells selectively and can protect non-transformed glial cells from death. These and other findings reviewed here might set the basis for a potential use of cannabinoids in the management of gliomas." Last year, a clinical review in the journal Nature Reviews Cancer made similar recommendations, noting that cannabinoids possess a "favorable drug safety profile" and have shown in clinical trials to inhibit various forms of cancerous tumors, including gliomas, lung carcinoma, breast cancer, skin cancer, thyroid cancer, lymphoma, and prostate cancer. Warning on Proton Pump Inhibitors and Fracture Risk

FDA is cautioning healthcare professionals and patients that proton pump inhibitors (PPIs) may increase the risk of fractures of the hip, wrist and spine. The drugs' labeling will be revised to reflect these concerns. PPIs are used to reduce gastric acidity, and include Nexium (esomeprazole), Dexilant (dexlansoprazole), Prilosec (omeprazole), Zegerid (omeprazole and sodium bicarbonate), Protonix (pantoprazole), Aciphex (rabeprazole) and Prevacid (lansoprazole). The new safety information is based on FDA's review of several epidemiological studies. These studies found that patients who received high doses of the drugs or took them for a year or more had the greatest fracture risk. Since most of these patients were older than 50, the increased risk was observed mainly in this age group. Healthcare professionals prescribing proton pump inhibitors should consider the possibility of an increased fracture risk, and whether a lower dose or shorter duration of therapy might be possible. Patients at risk for osteoporosis should have their bone status managed, and should supplement their diets with adequate amounts of vitamin D and calcium. Patients taking prescription PPIs should understand the possibility of an increased fracture risk, but they should be told not to stop their medication without consulting their healthcare professional. People using over-the-counter PPIs to treat heartburn should be cautioned not to take these drugs for more than 14 consecutive days, and not to <b>...</b>
