RhoG
RhoG (Ras homology Growth-related) (or ARGH) is a small (~21 kDa) monomeric GTP-binding protein (G protein), and is an important component of many intracellular signalling pathways. It is a member of the Rac subfamily of the Rho family of small G proteins and is encoded by the gene RHOG.
Discovery
RhoG was first identified as a coding sequence upregulated in hamster lung fibroblasts upon stimulation with serum. Expression of RhoG in mammals is widespread and studies of its function have been carried out in fibroblasts,leukocytes,neuronal cells,endothelial cells and HeLa cells. RhoG belongs to the Rac subgroup and emerged as a consequence of retroposition in early vertebrates. RhoG shares a subset of common binding partners with Rac, Cdc42 and RhoU/V members but a major specificity is its inability to bind to CRIB domain proteins such as PAKs.
Function
Like most small G proteins RhoG is involved in a diverse set of cellular signalling mechanisms. In mammalian cells these include cell motility (through regulation of the actin cytoskeleton),gene transcription,endocytosis,neurite outgrowth, protection from anoikis and regulation of the neutrophil NADPH oxidase.

What is Rh incompatibility? It occurs when a father who is Rh-positive and a mother who is Rh-negative have a baby that is Rh-positive. The baby's Rh-positive blood will leak into mom’s circulation (most likely during delivery), and this will cause the mother's to build antibodies.
The baby of the first pregnancy is not typically harmed. However, the baby during the second pregnancy, who is Rh-positive, will be affected because those antibodies created during the first pregnancy are in mom's circulation. These antibodies will attack the baby's red blood cells.
This will lead to hemolytic disease of the newborn and cause: anemia, jaundice, heart failure, swelling/edema, splenomegaly along with hepatomegaly.
The key is to keep mom from being sensitized, which is done through giving her a medication called Rhogam. This medication is given at 28 weeks and again at 72 hours after delivery if the baby is Rh-positive.

At the cellular level, the absence of ERI species impairs Ca2+ -mediated formation of adherens junctions, critical to maintain mechanical integrity in the epidermis. Our findings support a key role for ERI species in integrin-independent stabilization of the microtubule network in differentiated keratinocytes.

ELMO2 belongs to a family of scaffold proteins involved in phagocytosis and cell motility. ELMO2 can simultaneously bind integrin-linked kinase (ILK) and RhoG, forming tripartite “ERI” complexes. The latter are involved in promoting β1 integrin-dependent directional migration in undifferentiated epidermal keratinocytes. ELMO2 and ILK have also separately been implicated in microtubule regulation at integrin-containing focal adhesions. On differentiation, epidermal keratinocytes cease to express integrins, but ERI complexes persist. We now show an integrin-independent role of ERI complexes in modulation of microtubule dynamics in differentiated keratinocytes. Depletion of ERI complexes by inactivating the Ilk gene in these cells reduces microtubule growth and increases the frequency of catastrophe. Reciprocally, exogenous expression of ELMO2 or RhoG stabilizes microtubules, but only if ILK is also present. Mechanistically, activation of Rac1 downstream from ERI complexes mediates their effects on microtubule stability. In this pathway, Rac1 serves as a hub to modulate microtubule dynamics through two different routes: (1) Phosphorylation and inactivation of the microtubule destabilizing protein stathmin, and (2) phosphorylation and inactivation of GSK-3β, which leads to the activation of CRMP2, promoting microtubule growth. At the cellular level, the absence of ERI species impairs Ca2+ -mediated formation of adherens junctions, critical to maintain mechanical integrity in the epidermis. Our findings support a key role for ERI species in integrin-independent stabilization of the microtubule network in differentiated keratinocytes.

RhoG
RhoG (Ras homology Growth-related) (or ARGH) is a small (~21 kDa) monomeric GTP-binding protein (G protein), and is an important component of many intracellular signalling pathways. It is a member of the Rac subfamily of the Rho family of small G proteins and is encoded by the gene RHOG.
Discovery
RhoG was first identified as a coding sequence upregulated in hamster lung fibroblasts upon stimulation with serum. Expression of RhoG in mammals is widespread and studies of its function have been carried out in fibroblasts,leukocytes,neuronal cells,endothelial cells and HeLa cells. RhoG belongs to the Rac subgroup and emerged as a consequence of retroposition in early vertebrates. RhoG shares a subset of common binding partners with Rac, Cdc42 and RhoU/V members but a major specificity is its inability to bind to CRIB domain proteins such as PAKs.
Function
Like most small G proteins RhoG is involved in a diverse set of cellular signalling mechanisms. In mammalian cells these include cell motility (through regulation of the actin cytoskeleton),gene transcription,endocytosis,neurite outgrowth, protection from anoikis and regulation of the neutrophil NADPH oxidase.
