Sarin, or GB, is an organophosphorus compound with the formula [(CH3)2CHO]CH3P(O)F. It is a colorless, odorless liquid, used as a chemical weapon owing to its extreme potency as a nerve agent. It has been classified as a weapon of mass destruction in UN Resolution 687. Production and stockpiling of sarin was outlawed by the Chemical Weapons Convention of 1993 where it is classified as a Schedule 1 substance.
Production and structure
Sarin is a chiral molecule (typically racemic), with four substituents attached to the tetrahedral phosphorus center. The SP form (shown) is the more active enantiomer due to its greater binding to acetylcholinesterase. It is prepared from methylphosphonyl difluoride and a mixture of isopropyl alcohol.

Isopropylamine is added to neutralize the hydrogen fluoride generated during this alcoholysis reaction. As a binary chemical weapon, it can be generated in situ by this same reaction.
Biological effects
Its mechanism of action resembles that of some commonly used insecticides, such as malathion. In terms of biological activity, it resembles carbamate insecticides such as sevin and medicines pyridostigmine, neostigmine, and physostigmine. Like other nerve agents, sarin attacks the nervous system.
Specifically, sarin is a potent inhibitor of the enzyme cholinesterase. Sarin acts on cholinesterase by forming a covalent bond with the particular serine residue at the active site. Fluoride is the leaving group, and the resulting phosphoester is robust but biologically inactive. With the enzyme inhibited, acetylcholine builds up in the synapse and continues to act so that any nerve impulses are, in effect, continually transmitted. Normally, the acetylcholinesterase breaks down the acetylcholine in the synaptic cleft in order to allow the effector muscle or organ to relax...

Lessons Learned: Tokyo Sarin Gas Attack
On March 20, 1995, Aum Shinrikyo, a religious cult based in Japan, used chemical weapons in a terrorist attack on Tokyo's subway system. During the morning rush hour, five cult members carried bags of liquid sarin into Tokyo's subway and pierced the bags with the tips of their umbrellas, allowing the deadly nerve agent to evaporate and spread. The attack killed twelve people and injured thousands more.
James M. Lindsay, CFR's senior vice president and director of studies, argues that the 1995 sarin gas attack serves as a reminder that technology now "makes it possible for groups and individuals to carry out the kinds of attacks that once only government could undertake." Concerns over the spread of such technology, he says, lie at the heart of the current debate surrounding the manipulation of the H5N1 bird flu virus in laboratories. Such debates will continue to "grow more heated," he predicts, "as technological advances make it possible to do more and more with less and less training."

Nerve Agent History WWII
First discovered accidentally during the 1930s by industrial chemists in Germany conducting pesticide research, the nerve agents Tabun (GA) and Sarin (GB) were developed into chemical weapons and stockpiled by the Nazi regime. Fortunately, Hitler did not order their use during World War II because German intelligence believed--incorrectly--that the United States and the Soviet Union had developed similar weapons. After the war, the victorious Allies competed among themselves for the secrets of the Nazi nerve agent program. In the early 1950s, British industrial scientists accidentally discovered a second generation of nerve agents that were even more toxic than Sarin and were dubbed "V agents" because of their venomous (skin-penetrating) properties. During the Cold War, the United States and the Soviet Union pursued a chemical arms race in which they produced and stockpiled various nerve agents in the thousands of tons. For a detailed history for the general reader of the discovery, development, proliferation, and control of nerve agents such as Tabun, Sarin, Soman, and VX, read the 2006 book, War of Nerves: Chemical Warfare from World War I to Al-Qaeda, By Jonathan B. Tucker. During the 1960s, ocean dumping, openpit burning, and land burials were the U.S. Armys method of destroying chemical weapons. In 1969, the National Academy of Sciences recommended that ocean dumping be avoided. In the late 1960s President Nixon halts the production of chemical weapons. In 1972, the Army formed the U.S. Army Materiel Commands Program Manager for Demilitarization of Chemical Materiel, headquartered at Picatinny Arsenal, near Dover, NJ. The Army developed chemical weapons disposal methods using incineration and chemical neutralization. Project Eagle incinerates six million pounds of mustard agent and neutralizes eight million pounds of nerve agent GB (sarin) at Rocky Mountain Arsenal, Colo. between 1972 and 1976. Today, The U.S. Army's Chemical Materials Agency (CMA) safely stores and destroys the nation's aging chemical weapons and recovers the nation's chemical warfare materials. The agency is based at the Edgewood Area of the Aberdeen Proving Ground in Maryland, while other managers and staff fulfill the agency's mission from weapons storage and disposal at locations across the county. Syria has begun mixing chemicals that can be used to make deadly sarin gas, a U.S. official told AFP Monday, amid fears that President Bashar al-Assad's forces could attack rebels with chemical weapons.
"We've picked up several indications which lead us to believe that they're combining chemical precursors," the official said, on condition of anonymity, adding that the operation was apparently aimed at making sarin.
U.S. President Barack Obama directly warned Syria's President Bashar al-Assad Monday that he would face "consequences" if he made the "tragic mistake" of turning chemical weapons on his own people.
"Today, I want to make it absolutely clear to Assad and those under his command, the world is watching. The use of chemical weapons is and would be totally unacceptable," Obama said.
Earlier, a senior U.S. defense official said U.S. and allied intelligence have detected Syrian movement of chemical weapons components in recent days, the Associated Press reported, as the Obama administration strongly warned the Assad regime against using them.
A senior defense official said intelligence officials have detected activity around more than one of Syria's chemical weapons sites in the last week. The defense official spoke on condition of anonymity because he was not authorized to speak publicly about intelligence matters.
Secretary of State Hillary Rodham Clinton, in Prague for meetings with Czech officials, reiterated President Barack Obama's declaration that Syrian action on chemical weapons was a "red line" for the United States that would prompt action.
"We have made our views very clear: This is a red line for the United States," Clinton told reporters. "I'm not going to telegraph in any specifics what we would do in the event of credible evidence that the Assad regime has resorted to using chemical weapons against their own people. But suffice it to say, we are certainly planning to take action if that eventuality were to occur."
Syria said Monday it would not use chemical weapons against its own people. The Ministry of Foreign Affairs said Syria "would not use chemical weapons - if there are any - against its own people under any circumstances."
Syria has been careful never to confirm that it has any chemical weapons.
The use of chemical weapons would be a major escalation in Assad's crackdown on his foes and would draw international condemnation. In addition to causing mass deaths and horrific injuries to survivors, the regime's willingness to use them would alarm much of the region, particularly neighboring states, including Israel.
At the White House, press secretary Jay Carney said, "We are concerned that in an increasingly beleaguered regime, having found its escalation of violence through conventional means inadequate, might be considering the use of chemical weapons against the Syrian people. And as the president has said, any use or proliferation of chemical weapons by the Syrian regime would cross a red line for the United States. "

Washington, DC, United States (4E) -- A U.S. official has claimed that movements of chemical stockpiles in Syria indicate preparation of the deadly nerve gas sarin.
The anonymous official told Wired.com on Monday that the Syrian military are moving together stockpiles of sarin gas ingredients isopropanol, popularly known as rubbing alcohol, and methylphosphonyl difluoride, which were previously stored separately to prevent them from mixing and the accidental release of the nerve agent. The official also said the chemicals also have been moved in such a way that they can be loaded on a plane for dropping.
A CBS News report said commercial satellite photos of Syria's two dozens chemical weapons bases last week indicate that ingredients are being brought together.
Israel National News reported that Syria has 500 metric tons of isopropanol and methylphosphonyl difluoride stored separately. It is believed to make Syria the world's third-largest possessor of chemical weapons after the U.S. and Russia.
The claim heightened concern by Washington and Israel that chemical weapons might fall into wrong hands like the Hezbollah in Lebanon and other terrorist groups seeking the destruction of the Jewish state.
U.S. President Barack Obama had just warned Syrian President Bashar al-Assad that using chemical weapons will force the U.S. to intervene and send troops to Syria to secure such weapons. The U.S. military is also considering airstrikes on Syria's chemical bunkers to prevent Damascus from weaponizing chemicals.

Sarin, used in two terror attacks in Japan in the 1990s, is a man-made nerve agent which can cause convulsions, respiratory failure and death.
The intelligence appeared to explain a series of fresh warnings issued by Washington that the use of chemical weapons by the Assad government would cross a "red line" and invite unspecified US action.
